Factors affecting the measurement of atomic fluorescence spectrophotometer

At present, the analysis method of atomic fluorescence photometer has been widely used in health inspection, agriculture, metallurgy, geology, environmental protection, medicine and other fields, so the verification and calibration of atomic fluorescence photometer is an inevitable requirement for the measurement and testing industry to adapt to the development of analysis technology.
However, in the daily verification and calibration process, the results of technical indicators such as instrument sensitivity, precision and linearity may not be satisfactory due to the negligence of some common minor problems, which are briefly summarized as follows:
1. Room temperature
The speed of hydride reaction is affected by temperature, when the room temperature is lower than 15 °C, the diluted standard solution of arsenic and antimony should be placed for at least 30min. According to experience, the room temperature of the instrument is between (15~30) °C, subject to about 22 °C, and is relatively stable.
Second, the lamp position
The beams emitted by the two hollow cathode lamps should converge at the center of the flame of the atomizer quartz furnace to excite the resulting atomic fluorescence. When determining antimony and arsenic, the root of the flame should be used as much as possible. The position of the atomizer has a great influence on the sensitivity and stability of the instrument. The height of the atomizer is the furnace height, which refers to the distance between the mouth of the quartz furnace and the center of the photomultiplier tube, and the empirical recommendation value is 8mm. The furnace height increases, while the quartz furnace decreases.
Third, the negative high voltage and lamp current of the photomultiplier tube
Increasing the negative high voltage and lamp current can improve the sensitivity, but at the same time reduce the stability, so when the sensitivity can meet the requirements, the negative high voltage and lamp current should be reduced as much as possible. And choosing too large lamp current will shorten the life of the lamp, and in some cases may cause the bending of the working curve. Usually, the lamp current of arsenic lamp is adjusted to about 60mA, and the negative high voltage is placed around 300V.
Fourth, acidity
The hydride reaction occurs in an acidic medium, and the relative standard deviation measured by the instrument is less than 10% in the acidity range of 20%~5%. Due to the high sensitivity of instrument analysis, the requirements for the purity of the reagent during the determination are high, especially when the purity of the acid is not high, the blank value will be high and unstable, resulting in unsatisfactory linearity of the working curve.
Therefore, special attention should be paid to the use of high-grade pure hydrochloric acid, and at the same time use secondary deionized water to prepare the solution to reduce the blank.
In the verification and calibration of atomic fluorescence photometer, it is necessary to select the most suitable conditions according to the specific situation, and try to eliminate interference in order to make the instrument reach a good detection level.
